Results

This study enrolled a subgroup of 20 subjects from within a larger study comparing lithium (Li) and lamotrigine (LTG) for bipolar depression. FDG-PET data were obtained at baseline from 11 patients treated with Li and 9 patients treated with LTG. Eight normal controls were also scanned. Ten patients treated with Li were also scanned at 2 weeks, and 5 were scanned at 16 weeks. Nine patients treated with LTG were scanned at 2 weeks, and 6 were scanned at 16 weeks. Compared to normal controls, patients had lower activity in left middle frontal gyrus and left fusiform gyrus. Patients had increased activity in posterior hippocampus, medial postcentral gyrus, and cerebellum. Patients treated with Li showed an increase in activity in posterior cingulate and decreased activity in the right parietal lobe and the left precentral gyrus, compared to baseline. Patients treated with LTG showed an increase in activity in inferior temporal cortex and decreased activity in the left and right precentral gyrus, compared to baseline. Changed activity in left parahippocampal gyrus and posterior cingulate predicted response to LTG. Changed activity in left inferior parietal cortex and temporal cortex predicted a response to Li.
